Arrayit Microarray Stripping Buffers A, B and C (1L of 1X Solution) are an advanced 3-buffer set that allows DNA microarray stripping and reuse up to 10 times. Arrayit Microarray Stripping Buffers can improve the affordability of DNA microarrays for research, life sciences and testing applications.

Arrayit HTW capacity is 25 glass slides (1 x 25 x 76 mm) and 500 ml of each of the 1X solutions.

Short Protocol (Steps 1-7)
1. Transfer glass substrate slide microarrays to a High-Throughput Wash Station.
2. Wash 10 min at room temperature in 500 ml of Microarray Stripping Buffer A.
3. Transfer substrate slide rack to a second High-Throughput Wash Station.
4. Wash 10 min at room temperature in 500 ml of Microarray Stripping Buffer B.
5. Transfer substrate slide rack to a third High-Throughput Wash Station.
6. Wash 1 min at room temperature in 500 ml of Microarray Stripping Buffer C.
7. Spin glass substrate slides dry for 5 sec using Microarray High-Speed Centrifuge.
Complete Protocol (Steps 1-7)
1. Transfer glass substrate slide microarrays to a High-Throughput Wash Station. Arrayit High-Throughput Wash Stations Cat. HTW have a capacity for 25 glass microarray slides in the slide rack and 500 ml of 1X stripping buffer in the reservoir. Add 500 ml of Arrayit Microarray Stripping Buffer A to the HTW reservoir before adding the microarray slides. Make sure to keep the microarray slides hydrated at all times during the stripping steps as drying may elevate background fluorescence.
2. Wash 10 min at room temperature in 500 ml of Microarray Stripping Buffer A. Use a magnetic stir plate and moderate mixing during the stripping steps. Make sure to keep the microarray slides hydrated at all times. Arrayit HTW accommodates microarrays from Arrayit, Agilent and other manufacturers. Affymetrix customers can used the microfluidic station in conjunction with Arrayit stripping buffers for the stripping steps.
3. Transfer substrate slide rack to a second High-Throughput Wash Station. Quickly transfer the slides from the HTW containing 500 ml of 1X Arrayit Microarray Stripping Buffer A to a second HTW containing 500 ml of 1X Arrayit Microarray Stripping Buffer B.
4. Wash 10 min at room temperature in 500 ml of Microarray Stripping Buffer B. Moderate agitation on a magnetic stir plate should be used during this stripping step. Make sure to keep the microarray slides hydrated at all times. Arrayit HTW accommodates microarrays from Arrayit, Agilent and other manufacturers. Affymetrix customers can used the microfluidic station in conjunction with Arrayit stripping buffers for the stripping steps.
5. Transfer substrate slide rack to a third High-Throughput Wash Station. Quickly transfer the slides from the HTW containing 500 ml of 1X Arrayit Microarray Stripping Buffer B to a third HTW containing 500 ml of 1X Arrayit Microarray Stripping Buffer C.
6. Wash 1 min at room temperature in 500 ml of Microarray Stripping Buffer C. Moderate agitation on a magnetic stir plate should be used during this stripping step. Make sure to keep the microarray slides hydrated at all times. Arrayit HTW accommodates microarrays from Arrayit, Agilent and other manufacturers. Affymetrix customers can used the microfluidic station in conjunction with Arrayit stripping buffers for the stripping steps.
7. Spin glass substrate slides dry for 5 sec using Microarray High-Speed Centrifuge. Arrayit High-Speed Centrifuges Cat. MHC110V provide a rapid and convenient way to dry microarray slides after stripping. Make sure to insert the slide completely into the rotor and wear safety glasses at all times during use. A 5 second drying step should be sufficient to dry the microarray slide in preparation for the next round of hybridization.

- Designed to remove fluorescent probe molecules from microarrays post-hybridization
