- VacuSeal™ bags are reusable
- Accommodates 96-, 384- and 1536-well microplates
Safety and Maintenance
VacuSeal™ instruments should be operated on a sturdy and dry laboratory bench. Do not attempt to operate these instruments on wet or hot surfaces. Do not immerse these instruments or power cord in water or any liquid. Unplug before cleaning and use only mild soap or cleaners compatible with the instrument housing. Avoid contact with acetone and other strong solvents and acids or bases.
Short Protocol
- Connect the instrument to a voltage-appropriate electrical socket and make sure that the indicator light is illuminated.
- Seal the microplate with an adhesive foil seal (optional).
- Place microplate inside the VacuSeal™ Bag with the textured side down.
- Place the top edge of the bag inside the VacuSeal™ instrument.
- Press the vacuum button and press down on the lid.
- Wait 10-20 seconds until the VacuSeal™ instrument has completed the vacuum and sealing steps.
- Remove the vacuum-sealed microplate from the instrument and store at an appropriate temperature for your samples.
- Repeat the steps above to VacuSeal™ another microplate.
Complete Protocol
- Connect the instrument to a voltage-appropriate electrical socket and make sure that the indicator light is illuminated.
- Seal the microplate with an adhesive foil seal. Use of the foil seal is optional, however it will provide an effective barrier against cross contamination.
- Place microplate inside the VacuSeal™ Bag with the textured side down. The textured side down orientation will ensure that the instrument pulls a proper vacuum and creates a proper heat seal. If a foil seal is not being used, make sure not to invert the microplate. The 15 x 25 cm bags (Cat. BVS) accommodate a variety of 96, 384 and 1536 plate thicknesses and sizes. The larger 20 x 29 cm bags (Cat. BVS2) are also available to allow vacuum-sealing of larger items.
- The microplate and bag can be placed on a slide box in front of the VacuSeal™ machine to create the proper height for vacuum sealing. Using a slide box sets the proper level of the bag and microplate for easy operation.
- Align the open end of the bag inside the VacuSeal™ channel and make sure that there are no wrinkles in the bag along the seal bar. Failure to properly align the open end of the bag with the air channel will result in an inefficient vacuum sealing process.
- Press down on the lid gently until air starts to exit the bag, then release. Never attempt to disengage the VacuSeal™ instrument during operation as this can damage the instrument. Pressing the heat seal button during the vacuum step will halt the vacuum process and seal the bag. Manual heat sealing can be used in cases where a lower vacuum pressure seal is required, such as may be the case when sealing microarray slide boxes with glass slides and other fragile laboratory items.
- The VacuSeal™ instrument will automatically complete the vacuum and sealing step once the vacuum starts pulling.
- Remove the vacuum sealed microplate from the system and store at an appropriate temperature for your samples. The VacuSeal™ bag can withstand a variety of temperatures including freezing at temperatures as low as -80°C.
- Repeat steps 1-9 to vacuum seal another microplate. Bags can be opened with scissors and reused for convenience.
Troubleshooting Tips
Bag fails to seal
- Make sure that bag is lying flat against the instrument sealing bar
Machine fails to pull a vacuum from the bag
- Improper alignment of the open edge of the bag inside the unit or improper bag orientation (make sure textured side is facing down)
Figure 1. Shown are two types of microplates sealed with VacuSeal™ technology including a microplate with a foil seal (left) and two microplates with lids (right).
Figure 2. Arrayit VacuSeal™ technology (Cat. VMS110V) was used to vacuum seal a 7 x 7 x 7 cm open plastic box containing 250 grams of sodium chloride. Vacuum sealing can be used to prevent the accumulation of moisture in hygroscopic laboratory reagents.
